Haemoglobin Electrophoresis

Hemolystae is prepared from blood in EDTA,or citrate, or heparine.
Use fresh or refrigerate the sample.
Principle
First hemolysate from the EDTA blood is made and then it is run for electrophoresis where Hb is separated in different bands.One can find the presense of abnormal Hb .
Normal
•Hb A = > 95 %
•Hb A2 = 1.5 to 3.7 %
•Hb F = < 2 %
Pattern of electrophoresis
1.A > S = Sickle cell trait,Sickle a-thalasemia.
2.S,F and no A = Sickle cell anemia,Sickle-beta thalasemia.
3.S > A and F = sickle- beta thalasemia.
4.A > C =Hb C trait
5.C, F and no A = Hb C disease,HbC-beta thalasemia.
6.C > A = Hb C – beta thalasemia.
